Our research aims to decipher how metabolic alterations of macrophage orchestrate immune responses in chronic inflammatory diseases, with a special focus on metabolic syndromes (type II diabetes, insulin resistance and atherosclerosis) and cancer. We integrate transcriptomic, metabolomic and epigenomic approaches to identify the metabolic checkpoints and mechanisms underlying the regulation of macrophage activation and functions during disease progression, particularly in specialized microenvironment such as adipose tissues in diabetes mellitus, the arterial microenvironment in atherosclerosis and tumor microenvironment. Understanding the metabolic-immunity circuits of macrophage in the pathogenic microenvironment can pinpoint the key regulators contributing to the progression of these metabolic disorders. Our final goal is to formulate novel strategies targeting immunometabolic circuits in adipose tissue macrophages and atherosclerotic macrophages and ameliorate these chronic metabolic disorders.
